Collagen Antibody induced Arthritis (CAIA): preclinical model

 

The Collagen Antibody Induced Arthritis (CAIA) model is a rapid and synchronized alternative to the Collagen Induced Arthritis model. A cocktail of arthritogenic monoclonal antibodies to CII combined with a boost of LPS is used to induce arthritis. These arthritis inducing antibodies recognize a wide range of epitopes on the CII molecule and thereby allow formation of large immune complexes on the cartilage. Since these pre-formed antibodies are administered to the animals, the initial phase of the immune response involving T and B cell interaction is not present. This makes the model far more rapid, with onset within 48 hours of LPS boost and a total duration of as little as 10 days.

 The shorter model length presents great advanatages for researchers:

  • less compound required to run studies
  • Synchronicity between onset of disease in animals simplifies data analysis and allows for more direct comparison between test groups.
  • Susceptibility to various strains including normally CIA-resistant and transgenic strains
  • Time to market: multiple studies can be run in the time it takes to run 1 CIA so you can efficiently move your compound through pre-clinical efficacy stages.
  • Reduce development time by 30%
