in vitro screening of compounds for inflammatory activity

ImmuneProfiler™

Cell based assays are quick and provide the ability to interpret and predict many biological properties of a potential drug compound. MD Biosciences offers ImmuneProfiler as a series of successive in vitro assays beginning with a general screen to evaluate anti-inflammatory activity in cultured primary human peripheral blood mononuclear cells (PBMCs).

Following the results of this intial screen, our immunologists will design a series of in vitro assays that will profile the immunomodulatory acitivity or your compound. This can then lead to mode of action studies and target disease models using an efficient and focussed strategy.

These inflammatory screens can be as general as the PBMC assay stimulated with LPS detailed below, or alternatively more disease specific stimulants can be used. A more specific example would be he use of NLR ligands to induce the inflammasome, the multi-protein complex recently discovered to be involved in the initiation and progression of the inflammatory response in rheumatoid arthritis.

Human PBMC in vitro assay

PBMC's are stimulated with your choice of stimulants plus various concentrations of compound. Cytokine levels are then evaluated and provide information on which cell based assay will be used next.

LPS-stimulated cytokine release from human PBMC's

Human PBMC's from 1 donor were incubated in the presence or absence of 0.5 mg/mL hydrocortisone. LPS or media (+/- LPS) was added and cells were further incubated. Cell culture supernates were harvested and cytokines were assayed.
