Indirect COX-2 activity Assay: Preclinical Contract Research

Cyclooxygenase (COX) converts arachidonic acid to PGH2, the first step in the biosynthesis of prostaglandins, thromboxanes and prostacyclins. The inducible COX isoform, COX-2, is responsible for the biosynthesis of prostaglandins under acute inflammatory conditions and is the target of anti-inflammatory activity of non-steroidal anti-inflammatory drugs.

In the indirect COX-2 activity assay, COX-2 is induced in human PBMCs by stimulation with LPS. PGE2 production is used as a readout of COX-2 activity.

Figure: Indirect COX-2 activity assay. PBMCs were incubated with test compounds for 1 hour prior to LPS stimulation. Adter 24 hours of LPS stimulation, supernatants were collected and analyzed for PGE2. Mean values are shown. Error bars represent standard deviations.

Interesting Fact

Preclinical drug discovery and development outsourcing can lower development costs, decrease risk, speed up timelines and increase the confidence of potential partners.

Client Testimonial

“Among all the CROs that I have used over the years, including recently for inflammation, MD Biosciences has been one of the very best in terms of scientific knowledge, expertise, data quality, timelines, flexibility and personal contacts.”
