Cyclooxygenase (COX) converts arachidonic acid to PGH2, the first step in the biosynthesis of prostaglandins, thromboxanes and prostacyclins. The inducible COX isoform, COX-2, is responsible for the biosynthesis of prostaglandins under acute inflammatory conditions and is the target of anti-inflammatory activity of non-steroidal anti-inflammatory drugs.
In the indirect COX-2 activity assay, COX-2 is induced in human PBMCs by stimulation with LPS. PGE2 production is used as a readout of COX-2 activity.
