New antibodies to Citrullinated Protein.

Citrullination of proteins is a post-translational modification where arginine is converted into citrulline by a family of enzymes called peptidylarginine deaminases (PADs). Proteins are susceptible to citrullination during inflammatory processes, and an autoantibody response to citrullinated proteins is a hallmark of Rheumatoid Arthritis (RA).

Antibodies to Structural components of the ECM (proteoglycans and glycosaminogycans)

Immunohistochemical composition of neocartilaginous graft ECM. Sections have been labled (green) with antibodies toward structural components of the extracellular matrix, i.e. glycoaminoglycans and proteoglycans. Cell nuclei have been counterstained red with propidium iodide.

BC-3 and BC-13 monoclonal Antibodies: IHC staining of articular cartilage.

Immunohistochemical staining of human articular cartilage from a patient with osteoarthritis of the knee using monoclonal antibodies BC-3 and BC-14.

Osteoarthritis (OA) is a chronic and debilitating joint disease that is characterized by progressive destruction of cartilage wherease Aggrecan degradation is initially observed followed by overt collagen degradation.

In the figure below, sections from osteoarthitic human articular cartilage were either stained with (a) Toluidine Blue (b) monoclonal antibodies BC-4; anti MMP-generated neoepitope...DIPEN or (c) BC-13, anti-aggrecanase generated neoepitope ....NTEGE. Toluidine blue staining indicates loss of proteoglycan staining (blueness) in the fibrillated surface zone of the cartilage. Immunostaining with BC-4 shows staining in the mid-zones of the articular cartilage in the territorial matrix. In contrast, immunostaining with mAb BC-13 shows staining in the surface zone and uppoer mid-zone.

Detecting Small Leucine Rich Proteoglycan (SLRP) Core Protein Fragmentation in meniscus, knee and hip articular cartilage.

Using SLRP Antibodies for Western Blotting of tissue extracts.

Presented is identification of intact SLRP proteins and fragments in age-matched macroscopically normal (N), osteoarthritic (OA) or fibrillated (F) knee articular cartilage (AC). Affinity-purified anti-caroxyl-terminal SLRP antibodies (PR-84), PR-85, PR-184) and PR-353) and a monoclonal antibody to full-length keratocan core protein (KER-1) to perform western blots of samples. The brackets above the lanes indicate that macroscopically normal and fibrillated articular cartilage were samples from the same individual monoarthritic joint for comparison.

Identification of SLRP core protein fragmentation in meniscus, knee and hip articular cartilage.

Presented is indentification of SLRP core protein fragmentation in 4M GuHCl extracts of meniscus, knee and hip articular cartilage of individual tissue specimens from total knee or hip replacement patients. The ages (years) of each specimen art indicated at the top of each lane.
